The European champions, who are among the favourites to claim a second World Cup title, eased to victory with Mikel Oyarzabal and Pedri scoring in the first half before a Pedro Gallese own goal.
It took Spain just two minutes to go ahead in Puebla, Oyazarbal firing his left foot shot from outside the box into the bottom corner.
Oyarzabal went close again, firing over after being set up by Pedri who added the second after 32 minutes.
Ferran Torres found his Barcelona colleague in the box and Pedri was able to fire in from close range.
Peru had several chances before the break, but were unable to cut the gap and Gallese turned the ball into his own net early in the second half as Spain coach Luis de la Fuente changed his entire line-up.
Injured trio Lamine Yamal, Nico Williams and Victor Munoz played no part in the match.
Peru pulled one back when Jairo Velez latched onto Marcos Lopez's through ball and slotted it past Arsenal's David Raya.
